چکیده

Maize occupies a significant place in the world agriculture. Yellow kernel maize contains mainly non-provitamin A carotenoids: lutein and zeaxanthin. The accumulation of provitamin carotenoids is regulated by favourable alleles lcyE crtRB1 genes could be used for enhancement these grain through breeding. In this study, molecular screening Romanian germplasm was performed, looking genes, level determined few selected lines. number 2746 inbred lines from seven research stations were subjected to PCR amplification order identify alleles. It 27 carrying nine with unfavourable (four groups total), which total carotenoids, lutein, zeaxanthin, β-cryptoxanthin, β-carotene retinol equivalents HPLC. Out analysed, 23.53% contained one or both allele gene most widespread (584 lines), followed (55 while detected only 7 Inbred showed highest levels those high β-cryptoxanthin; had 60% higher than two

Maize (Zea mays L.).

Agrobacterium tumefaciens-mediated transformation is an effective method for introducing genes into maize. In this chapter, we describe a detailed protocol for genetic transformation of the maize genotype Hi II. Our starting plant material is immature embryos cocultivated with an Agrobacterium strain carrying a standard binary vector. In addition to step-by-step laboratory transformation proced...

Association Mapping for Enhancing Maize (Zea mays L.) Genetic Improvement

Association mapping through linkage disequilibrium (LD) analysis is a powerful tool for the dissection of complex agronomic traits and for the identifi cation of alleles that can contribute to the enhancement of a target trait. With the developments of high throughput genotyping techniques and advanced statistical approaches as well as the assembling and characterization of multiple association...

Lignification of developing maize (Zea mays L.) endosperm transfer cells and starchy endosperm cells

Endosperm transfer cells in maize have extensive cell wall ingrowths that play a key role in kernel development. Although the incorporation of lignin would support this process, its presence in these structures has not been reported in previous studies. We used potassium permanganate staining combined with transmission electron microscopy - energy dispersive X-ray spectrometry as well as acrifl...

CO(2) Compensation Concentration in Maize (Zea mays L.) Genotypes.

Heichel and Musgrave (4, 5) reported photosynthetic rates of maize genotypes ranging from 21 to 85 mg CO2 dmhr-' which were inversely associated with similar large differences in CO2 compensation concentration of from 9 to 25 plll. Since genotypic differences of the magnitude reported by Heichel and Musgrave would be of great value in breeding for increased photosynthetic efficiency, we conduct...

Alterations in Carbohydrate Intermediates in the Endosperm of Starch-Deficient Maize (Zea mays L.) Genotypes.

Metabolite levels in kernels of selected starch-deficient mutants of maize (Zea mays L.) were investigated to gain insight into partitioning of carbohydrate metabolism during kernel development. Several free sugars, hexose phosphates, triose phosphates, fructose-2,6-bisphosphate, and pyrophosphate were measured in normal, shrunken, shrunken-2, amylose extender dull waxy, and brittle genotypes, ...
